| Research Abstract |
(1) We have purified botrocetin from the snake venom of Bothrops jararaca. Then, we obtained various anti-botrocetin monoclonal antibodies by immunizing the purified botrocetin to Balb/c mice.
(2) Among various anti-botrocetin monoclonal antibodies, we obtained the antibody, BCI-7, which completely blocks von Willebrand factor (vWF) binding to botrocetin.
(3) By immuno-histochemical studies using the BCI-7 antibody, we found the positive cross-reactive substances in vascular subendothelium of human umbilical cord or placental tissues. Tissue specificies or fashion of these findings are now under way.
(4) With regard to the initiation mechanism of vWF-glycoprotein (GP) Ib interaction, we obtained new findings that, under rheological situations, conformational changes of vWF upon its immobilization are not responsible for the enhanced GP Ib-binding ability (Blood, 88 : 3854-3861,1996).
