Research Abstract

At first, using immunoblot and ELISA,the incidence of anti-carbonic anlydrase (CA) antibodies in Rheumatic autoimmune disease (RAD) was investigated. Anti-CA antibodies were found in 20-30% of sera from patients with RAD.All anti-CA sera were reactive with both human CAI and CAII.The correlation of it with clinical syndrome has been investigated. Second, epitopes on CAI and CAII antigen which recognized by RAD sera or natural autoantibody found in healthy individuals were analyzed using synthesized peptides. Anti-CA antibodies in RAD patients recognized alfa-helical portion on amino-terminus of CA and active domains of CA enzyme. This may evident the inhibitory effect of anti-CA antibodies against CA activity which previously discussed. In contrast, natural autoantibodies exclusively reacted with a portion located at carboxy-terminus of CA.These observation may suggest the distinguished mechanism of autoimmunity in RAD.We also reported a patient with anti-CA antibody who has similar symptoms to CA deficiency.
