Research Abstract |
In the administration of anti-cancer drugs, it is very important if the drugs can reach to the cancer cells. This permeability of drug to cancer cells might be much influenced by the distribution of micro vessels in the tumor tissue. For the purpose of investigating these relationship, we have done the following experiments. In fundamental study with murine tumor tissue, the vessel density after radiation therapy was investigated and was compared to the drug distribution in the tumor after i. v. injection. The tumor vessel density started to decrease 3 days after irradiation. This was accompanied by the decrees of drug distribution. However, the densoty recovered within 14 days after irradiation with the improvement of drug distribution. In the clinical study, the resected cancer tissues from tongue cancer patients were used to study the distribution of proliferating cells and PAI,UPA positive cells. The ratio of PCNA,UPA and PAI-1 positive cells were compared to the patient's survival. The treatment results of the patients with moderate PCNA and PAI-1 positive cell ratio were worse than the other group. The ratio of UPA positive cell did not influenced the patients survival. There was a strong positive relationship between the PCNA and PAI-1 positive cell ratio. Although, the mechanism of this positive relations and the may in which the PCNA and PAI-1 positive cell ratio influence the survival was unclear.
