Research Abstract

我々はヒト破骨細胞様細胞のcDNAライブラリーから破骨細胞形成を促進する因子としてAnnexinIIを同定した(J.Biol.Chem.269:28696,1994)。AnnexinIIは骨髄細胞からの破骨細胞形成を促進し骨吸収促進作用も示した。一方、従来細胞質内蛋白と考えられていたAnnexinIIがラット乳癌細胞を含む種々の細胞の細胞膜外に発現され、細胞外マトリックス、内皮細胞等への接着に関与していることも明らかになっている。そこで我々は乳癌細胞がAnnexinIIを発現して破骨細胞形成を促進し骨転移病巣の形成を促している可能性を検証するために以下の検討を進めた。1)乳癌細胞株がAnnexinIIを発現することを、mRNAレベル(AnnexinII cDNAによるNorthern blot)及び蛋白レベル(抗AnnexinII抗体によるWestern blot,Flowcytometry)で確認した。2)マウス乳癌細胞とマウス骨髄細胞の共存培養の系を確立し、AnnexinIIの発現と破骨細胞形成促進能の相関の検討を行い、さらにAnnexinIIのsense-,antisense-plasmidを乳癌細胞に導入したサブクローンを確立し、共存培養における破骨細胞形成促進能を検討した。マウス乳癌細胞MMT060562はサイトカイン非存在下で骨髄細胞による破骨細胞形成を促進するが、MMTにAnnexinIIを強発現させた細胞株は有意に強い破骨細胞形成刺激効果を示し、軽度AnnexinIIを抑制した細胞株は、破骨細胞形成促進が弱い傾向があった。更にin vivoでの骨転移形成能の検討を予定している。3)ヒト乳癌の臨床サンプルを集積し、乳癌原発巣でのAnnexinIImRNAの発現を確認し、乳癌原発巣および骨転移巣サンプルでのAnnexinII発現について免疫組織学的検討を行っている。
