Research Abstract |
9. Rat pancreatic islets were isolaed using the collagenase digestion techniqe. And they were dispersed by pipetting to small clsters of islet cells that each contained approximately 10 to 50 cells. Cells were loaded with fura-2 acetoxymethylester for 30minutes and intracellular calcium were measured over the clusters of beta cells. The elevation of intracellular calcium in one cell of the cell cluster in response to high conentration of glucose spreads to the neighboring cells by the gap junction in various manners. In one case, the wave of high intracellular calcium concentration originated from one cell spreads in various directions concentrically. In another case, it spreads in the only one specific direction. It is speculated that as for the conductivities of the calcium waves, there are different kinds of beta cells as if the conduction system for the high concentration of calcium waves might exist. Next, we also used the clusters of pancreatic beta cells from the streptozotocin-induced NIDDM model rats and analyzed the spreading of the calcium waves in responese to the high glucose atimulation. Although the number of the beta cells was not enough for the analysis due to the diabetogenic effect of streptozotocin, there seems to be an impairement of the spreading of the calcium waves among the beta cells.
