Research Abstract |
We examined the effects of 8-chloro-adenosine 3' : 5'-monophosphate (8-Cl-cAMP), site-selective analogue of cAMP,and its metabolite 8-chloro-adenosine (8-Cl-ad) on the proliferation of human adenocarcinoma cell line HSG cells and its derivative cell line HSG-AZA1, HSG-AZA3, and human adenoid squamou carcinoma cell line TYS cells in culture. Cultivation of these cells in thepresence of 8-Cl-cAMP (0.5muM) or 8-Cl-ad (lmuM) resulted in the growth inhibition and reduction of colony formation rates on plastic surface or in soft agar to under 10%. Whereas the morphological alteration of treated cells were not shown. Then, it was confirmed that the HSG cells treated with 8-Cl-cAMP (0.5muM) or 8-Cl-ad (1muM) were not induced apoptosis, because DNA pattern was similar to untreated cells in agar gel electrophoresis. In addition, the cAMP-dependent protein kinase A,isolated from treated HSG cells, were decreased.
