下垂体腺腫の細胞起源と遺伝子異常に関する研究

Research Abstract

凍結保存してあった10例の下垂体腺腫組織(成長ホルモン産生腺腫5例、プロラクチン産生腺腫5例)より、proteinase K-phenol-chloroform法を用いてDNAを抽出した。今回はMEN-1の遺伝子のallelic deletionの有無を検討し、成長ホルモン産生腺腫についてはGsα遺伝子のmutationの有無を検討した。MEN-1とGsα遺伝子の遺伝子の特定の部位をpolymerase chain reactionにより増幅し、増幅された遺伝子産物をpolyacrylamide gelで電気泳動し、single strand conformation polymarphism(SSCP)法により解析し、allelic deletionの有無を検討した。同時に免疫組織化学染色やin situ hybridization法により、下垂体腺腫の内分泌能についても検討した。今回の検討では、いずれの腺腫においてもこれらの遺伝子の変異は認められなかった。従ってその内分泌能との相関については明らかな知見は得られなかった。今後はGH、PRL、TSHの転写因子として働くPit-1の発現を免疫組織化学染色やin situ hybridization法により検討し、個々の腺腫の内分泌能との関連について調べたいと考えている。
