Research Abstract |
I HAVE SHOWN THAT IMMUNIZATION WITH A PEPTIDE TERMED RHCA 67-16 SPECIFIED BY RNA COMPLEMENTARY TO THAT ENCODING ACHR 61-76 CAN INDUCE POLYCLONAL AND MONOCLONAL ANTI-IDIOTYPIC (ID) ANTIBODIES (AB) WHICH RECOGNIZE THE PARATOPE OF ACHR 61-76 SPECIFIC AB AND ALSO BLOCK THE DEVELOPMENT OF EAMG.SINCE THE PRODUCTION OF SUCH DISEASECAUSING ACHR-SPECIFIC AB IS T-CELL DEPENDENT,WE HAVE TESTED WHETHER A PEPTIDE (DENOTED RHCA 611-001) SPECIFIED BY RNA COMPLEMENTARY TO THAT ENCODING A T CELL EPITOPE (ACHR 100-116 IN LEWIS RAT EAMG) COULD INDUCE AB WHICH RECOGNIZED RECEPTORS ON T CELLS (TCR) SPECIFIC FOR ACHR 100-116 AND WOULD THUS ALSO BLOCK THE DISEASE.POLYCLONAL AND MONOCLONAL (MAB) INDUCED BY IMMUNIZATION WITH RHCA 611-001 SPECIFICALLY BLOCKED THE PROLIFERATION OF TORPEDOACHR-SENSITIZED LYMPHOCYTES,STIMULATED WITH ACHR 100-116. FURTHERMORE A MAB AGAINST RHCA 611-001 ALSO IMMUNOPRECIPITATED A CELL SURFACE PROTEIN WITH THE CHARACTERISTICS OF A TCR FROM A ACHR 100-116 SPECIFIC T CELL LINE.ANALYSIS OF VB UASAGE OF EAMG SHOWED A LIMITED HETEROGENITY (VB 15) IN EAMG.SOME OF THE CLONED HAD A HIGH HOMOLOGY SEQUENCE BETWEEN THE CDR 3 REGIN OF TCR AND RHCA611-001.
