Research Abstract |
The mononuclear cells in cord blood were studied as a source of liver support unit. Initially most effective method of high yield on CD34 positive cells was discussed. The choice of anti-coaglant agent was the most important factor on the recovery of CD34 positive cells. Heparin was not effective because its short anti-coaglant activities. Thus the ACD solution was recomended for harvesting coad blood. The serum level of HGF were 0.494<plus-minus>0.117ng/ml in cord blood (n=4) and 0.358<plus-minus>0.047ng/ml in peripheral blood (n=4). The released HGF levels in the culture medium (n=5) with cytocaines were 0.126<plus-minus>0.094ng/10^6 cells on 24 hours and 0.448<plus-minus>0.424ng/10^6 cells on 72 hours. And without cytocaines (n=5), the levels were 0.125<plus-minus>0.080ng/10^6 cells and 0.152<plus-minus>0.121ng/10^6 cells, respectively. The calculated HGF release values of CD34+CD33-cells with cytocaines were 13.3<plus-minus>11.3ng/10^6 cells on 24 hours and 41.2<plus-minus>31.7ng/10^6 cells on 72 hours.
