Research Abstract

1.α位不斉炭素原子に水素原子を有する酸ハライドの塩基存在下のエステル化反応では,100%ラセミ化するという「化学の常識」を加熱活性化した塩基性アルミナを用いて打破し,窒素原子を保護した天然型L-α-アミノ酸を,ラセミ化することなく応用範囲の広いその第三級ブチルエステルに導いた。
2.前項を利用して,L-プロリン第三級ブチルエステルを合成,更にクマリン骨格と縮合させ,キラルアルコール,アミン類に対する光学純度検定に有用な^1H NMR専用のキラル誘導化試薬略号,CDA),クマリノプロリンの開発に成功した。
3.前項のクマリノプロリンでは「歯がたたない」立体障害の大きなキラルアルコール,アミン類に対しては,代りにより有用なクマリノピログルタミン酸を新たに開発,公表した。驚ろくべきは本化合物の化学反応性で,^1H NMR文献上皆無に近いキラル第三級アルコールのCDAとして利用可能であることが判明した。
課題研究の拡大として,L-α-アミノ酸のみならずキラルα-オキシカルボン酸とクマリン骨格を縮合させた。100%光学的に純粋なクマリノオキシカルボン酸を合成し,これらの化合物が又キラルアルコール,アミン類に対するCDAとして有用であることを実証し,報告した。
