In this study, in order to investigate how cells recognize biomaterials, mRNA that was expressed in attached HeLa S3 cells on various substrates was evaluated. The expressed HSP 70A,70B,90 and 47 mRNA were then isolated and detected using the RT-PCR method. As a result, 70B,90 and 47 mRNA expressions varied with differences in the hydrophilicity-hydrophobicity of the substrates. The HSP mRNAs expressions were largely induced in HeLa S3 cells that had adhered onto hydrophilic surfaces. On the other hand, however, in the cells that had adhered onto hydrophobic surfaces, the HSP mRNAs expressions were low. In the non-adhered HeLa S3 cells the tendency of HSP 70B and HSP 47 mRNAs expressions were the same as in those of adhered cells, while there were no significant differences in the HSP 90 mRNA expression among the samples. It is clear that the study of HSP mRNAs expressions is very useful in the evaluation of cell-polymer interactions. The HSP 70B mRNA expression could be a very sensitive marker for the investigation of cell-polymer interactions. HeLa S3 cells that adhered onto hydrophilic surfaces suffered a heat-shock like stress-response, whereas the cells on the hydrophobic surfaces did not suffer any stimulation. We would emphasize that the investigation of mRNA expression is a powerful tool for studying cell-polymer interactions or the evaluation of the biocompatibility of biomaterials.
