Research Abstract

本研究の目的は開発途上国の獣医臨床現場においても実用可能な高感度・簡便・迅速原虫病診断法を開発することである。本年度はタイレリアパルバとトキソプラズマについて特異的LAMP法を開発し、論文発表した(Zhang et al.2009、Thekisoe et al.2010)。タイレリアパルバは東アフリカ地域で流行している東海岸熱の病原原虫であり、ウシに重篤な疾病を引き起こす原虫として日本においても家畜伝染病に指定されている重要病原体である。一方、トキソプラズマは人獣共通感染症であり、こちらもタイレリアパルバ同様家畜衛生・公衆衛生上極めて重要な原虫である。今回開発したタイレリアパルバ検出用LAMP法では以前に本研究プロジェクトで開発し、論文発表した(Thekisoe et al.2007,Onderstepoort J.Vet.Res.74:339-342)HSP70遺伝子に対するLAMP法より優れた特異性と感度が達成できた。加えて今回開発したタイレリアパルバPIMおよびP150遺伝子に対するLAMP法は南アフリカ、ブルンジ、ルワンダ、タンザニア、ケニア、ウガンダで分子した原虫株DNAに対して良好な増幅を示したことから、本LAMP法はタイレリアパルバ野生株の検出に広く応用できる可能性が示唆された。将来的には現在研究が先行しているアフリカトリパノソーマフィールド診断用LAMPキット開発の経験を生かして、タイレリアパルバ等さまざまな原虫病検出用迅速診断キット開発に発展させたい。
