精子と卵子の起源を探る:性染色体ゲノム比較による卵生殖の進化の解明

Research Abstract

本研究は、高等動物や陸上植物をそれぞれ含む系統群ではほぼ不可能であるとえられる「雌雄二極化の源」を解明するための唯一のアプローチであるといえる。なぜなら、同型配偶から異型配偶・卵生殖までの進化を段階的に橋渡しする汎系統的な分子レベルの比較研究は、群体性ボルボックス目における優性生殖過程の観察と分子系統解析以外ほとんど存在しない。本研究においてはまず、群体性ボルボックス目緑藻において、クラミドモナスのような一方向的接合突起型同型配偶・単細胞緑藻とボルボックスのような卵生殖・群体性緑藻の中間段階にあると考えられる、両方向的接合突起型同型配偶・群体性緑藻ゴニウムと異型配偶・群体性緑藻プレオドリナの間で性染色体領域配列を決定、クラミドモナスおよびボルボックスの性染色体ゲノム配列データと比較する。
Gonium pectorale,Yamagishiella unicocca, Pleodorina starriiについてはクロモソーマルウォーキングとショットガンシーケンシングが継続中である。現時点で、G.pectorale minusでは450kb、plusでは300kb、Pl.starrii maleでは300kb,femaleでは200kbの配列が決定されている。また計画通り、同型配偶Yamagishiella unicoccaのマイナス型において、性決定遺伝子領域をターゲットとするプローブであるMID遺伝子配列を用いてクロモソーマルウォーキングとショットガンシーケンシングが継続中であり、現在までにY.unicocca minusで150kbの領域が配列決定済である。
われわれが卵生殖Volvox carteriで探索し配列決定に成功した性決定遺伝子ホモログVcMIDは米国の共同研究者らと共著の論文として掲載された(Ferris et al.Science印刷中。著者15名中、申請者は第12著者)。
