Research Abstract

昨年度に引き続き、森島らは、いくつかのアミノ酸置換西洋わさびペルオキシターゼ(HRP)を作製し、ペルオキシターゼ反応における近位側芳香族アミノ酸残基の役割について検討を行った。本年度は、近位側にチロシン残基を導入したHRPの2種の変異体F179Y(Phe179-〉Tyr)、F221Y(Phe221->Tyr)を作製し、プロスタグランジン合成酵素(PGHS)でみられるような反応中間体としてのチロシンラジカルが生成するかどうか検討した。その結果、Phe179位にチロシン残基を導入した変異HRPはそのESRスペクトルから、PGHSでみられるようなチロシンラジカルを生成していることが明らかになった。一方、Trpの導入でトリプトファンラジカルが生成したPhe221位では、一般に酸化され易いとされているチロシン残基を導入してもポルフィリンラジカルしか生成せず、このことは、蛋白質中のアミノ酸残基がラジカルになるかどうかは、アミノ酸固有の性質やポルフィリンからの距離だけではなく、そのアミノ酸の置かれた環境が大きく影響するものと考えられた。渡辺は、ヘム近傍のアミノ酸を置換したミオグロビンを種々のヘム酵素のモデルとして、用い、ミオグロビンにおいてもヘム周辺に適切なアミノ酸置換を行うことで、ペルオキシターゼやチトクロムP450と同様な反応中間体や触媒活性が実現できることを明らかにした。長谷は、いくつかのアミノ酸置換亜硫酸還元酵素を作製し、その基質認識機構や還元機構を検討した。城は一酸化窒素還元酵素の高分解能のX線構造解析と、そのイソシアニド複合体の可視吸収・赤外分光測定を用いて、一酸化窒素還元の分子機構を検討した。郷はペルオキシターゼなど多くの金属結合蛋白質のモジュール構造を明らかにすることにより、金属結合部位を形成する普遍的なモジュールを推定した。
