Research Abstract

CD8+キラーT細胞より認識されたHLA-A31により提示されるヒト胃癌拒絶抗原のnatural antigenic peptideと思われる分子を単離した。
1.一次構造はTys-Ser-Trp-Met-Aap-Ile-Ser-Cys-Trp-Ileの10ヶのアミノ酸よりなり(F4.2と命名)、N末3番目、6番目はA31結合モチーフと合致したアミノ酸であり、これらがアグレトープとしてA31に結合する主たるアミノ酸と推測された。
2.C末より2ヶアミノ酸を除くとキラーT細胞は反応せず、従って、N末より9番目のTrpがT cell epitopeと推測された。
3.この抗原は他の胃癌(MKN28)にもHLA-A31を発現させると拒絶抗原として働いた。しかも、この構造はF4.2と類似したものであることが推測された。従って胃癌ワクチンとしての可能性が考えられた。
