Research Abstract

本研究は、癌の新しい治療戦略の開発を目指した基礎研究であり、本年度は以下の成果を得た。
(1)アポトーシス誘導システムの開発:
これまでに、Fas受容体のデスドメインとエストロゲン受容体のエストロゲン結合領域の融合蛋白質を介して、エストロゲン依存性にアポトーシスを誘導するシステムの開発に成功してきた。このシステムは、特に肝癌・膵癌細胞に効率よくアポトーシスを誘導することが判明した。種々の癌細胞で、上記システムの感受性を検討したところ、fasのデスドメインに誘導されるアポトーシスシグナルに抵抗性を示す癌細胞の存在が明らかになったが、そのような細胞でも、ポリグルタミンを発現させると、アポトーシスが効率よく誘導されることを明らかにした。ポリグルタミンは、我々が、デスドメイン以外のアポトーシス誘導物質として、神経細胞のアポトーシスの解析から見いだしたものである。
(2)癌細胞特異的な遺伝子発現システムの開発:
前立腺癌に特異的に外来遺伝子を発現させる目的で、前立腺特異抗原(Prostate Specific Antigen)の遺伝子を単離し、前立腺、特に前立腺癌で特異的に遺伝子発現を誘導する遺伝子領域を同定することに成功した。この領域の支配下に転写活性化因子を発現させることによって、前立腺癌での特異性を維持したまま、外来性遺伝子を大量に発現させるシステムを構築する事ができた。一方、肝癌特異的に遺伝子を発現させるためにalpha fetoprotein等の肝癌に特異的に働くと報告されている遺伝子領域を単離し、実際にこれらの領域を使って、肝癌特異的な遺伝子発現系の構築も進行中である。
