Research Abstract

前年度に引き続き、リンパ球の分化と活性化、レパートリー形成について研究を進め、以下のような成果を得た。(1)低分子G蛋白Rap1がT細胞レセプターの刺激により活性化されインテグリンの活性化を誘導すること、さらにその遷延性の活性化はT細胞応答性と抑制し免疫記憶成立障害に至ることを示した。(2)CTLA-4が、SHP-2によらない新しいシグナル経路でT細胞のIL2産生を抑制することを明らかにした。(3)胚中心におけるB細胞に強く発現される新しい遺伝子GANPを同定し、これがDNA複製に関与するMCM3と結合することを明らかにし、B細胞のクローン増幅に関与している可能性を示した。(4)単一前駆細胞からのリンパ球の分化のアッセイ法を確立し、これを用いてT前駆細胞が胸腺内でT細胞レセプターβ鎖の再構成に先だって著明な細胞増殖を示しレパートリー拡張に寄与していることを示した。(5)プレB細胞レセプターのシグナルによって、免疫グロブリン遺伝子のH鎖の再構成の抑制(対立遺伝子排除)とL鎖遺伝子再構成の亢進が協調的に誘導されることを明らかにした。(6)NotchのリガンドDelta系の分子Pref-1が胸腺細胞の生存と増殖に関与していること、それはHes-1転写制御因子の発現誘導によっていることを示した。(7)IL7によって活性化されるStat-5が、CBPやp300と協調的に働くことによって胸腺細胞のT細胞レセプターγ鎖のgermline転写を誘導し、V-J遺伝子組み換えを引き起こしてγδ系列のT細胞分化を誘導することを明らかにした。
