Research Abstract

1.TGF-βはBリンパ球に作用してIgAの産生を促進する。一方PEBP2ファミリーの転写因子のうちPEBP2αCもBリンパ球の分化に関与している。本研究でSmad3とPEBP2αCが細胞内で結合することを示した。またIgCαのプロモーター領域にPEBP2とSmadの結合領域があり、Smad3がPEBP2αCと協調して転写活性を促進し、TGF-βの核内のターゲットとしてPEBP2が作用していることを明らかにした。
2.IL-6依存性に増殖するハイブリドーマMH60細胞にBMP2を添加するとIL-6存在下にアポトーシスによる細胞死を誘導することが明らかとなった。このアポトーシスはTAK1キナーゼの上昇の結果、p38MAPキナーゼが活性化されるためであることが確認された。また抑制型SmadであるSmad6はBMP2によるアポトーシスを抑制した。
3.メモリーB細胞の分化に関与する転写因子BCL6の活性がヒストン脱アセチル化酵素(HDAC)の阻害剤であるトリコスタチンAによって消失することを明らかにした。BCL6はHDACをリクルートし、BCL6を発現している細胞にトリコスタチンAを加えるとアポトーシスが起こらなくなることから、HDACを介してメモリーB細胞にアポトーシスを誘導していることが明らかとなった。
4.サイトカインの機能を負に調節する因子CIS1のトランスジェニックマウスを作成した。このマウスのリンパ球はIL-2の反応が弱いなどさまざまな表現型を示し、STAT5aとSTAT5bのノックアウトマウスと極似しており、CIS1がSTAT5の作用を個体レベルで抑制していることが証明された。
