Research Abstract

細胞膜のグローバルな構造形成と機能の研究が、本格的に始まりつつある。例えば、細胞膜のドメイン構造の形成、細胞全体の形態の形成と変化、細胞運動、細胞分裂、エンドサイトーシス、隣接細胞との相互作用のための構造の形成、膜受容体と細胞内情報伝達分子の集合と局在化、などの、細胞膜が大きな構造体として関与する重要な現象について、それらの分子機構を解明するための研究である。このためには、従来の膜に関するアプローチだけでは不十分であり、膜ドメインの形成と膜骨格/細胞骨格との相互作用を常に視点にいれておくことが必要である。一方、顕微鏡下で、機能している1分子、生細胞内での1分子を見て操作する技法の最近の進展は著しく、細胞膜と細胞骨格の相互作用が直接的に1分子のレベルで調べられるようになってきた。分子同士を押し付けたり、1分子を引っ張ってみたり、捻ってみたりすることで、初めて、分子間の相互作用が理解できるようになる。
本研究では、このような研究の発展段階に鑑み、細胞膜研究のエクスパートに、細胞骨格系の研究者と、1分子直視・操作の方法を開発している研究者を加えて、「膜タンパク質・脂質と膜骨格/細胞骨格との相互作用を直接に調べ、細胞膜のグローバルな構造形成と機能発現の機構を解明する」、という課題に答えられる新しい研究の方法と戦略を創出することを目指した。そのために、異分野の研究者によるパイオニア的共同実験と討論を実行し、1分子直視・操作の技術のさらなる開発と細胞膜系への応用法の開発を推進した。さらに、実際に、細胞膜でのドメイン構造形成の機構などを解明するための研究をおこなった。さらに、1原子レベルでの、膜分子間、及び、膜分子と膜表在性タンパク質との相互作用のパイオニア的研究もおこなった。
