Research Abstract

小腸からクローニングされたAquaporin(AQP)1、AQP3およびAQP4の各々の水チャネル分子を発現させたアフリカツメガエル卵を用いた実験では、コレラ毒素の添加によりAQP3の水透過性が抑制され、逆にAQP4の水透過性は促進された。AQP1の水透過性は影響を受けなかった。この結果を追試で確認するとともに、アデニル酸シクラーゼの促進剤であるforskolinの添加によってもコレラ毒素と全く同様の効果が得られることも確認した。これによってコレラ毒素のAQPに対する効果が細胞内cAMP濃度の上昇と関与していること、すなわち従来考えられているコレラ毒素の下痢発症メカニズムと同じ作用による可能性が高いことが示唆された。
また詳細なRT-PCRにより、AQP1、3、4以外のAQPのスクリーニングを小腸を材料に行ったところ、AQP7cDNAを新たにクローニングできた。またこれによりその他のAQPすなわちAQP0、2、5、6、8、9は小腸では発現していないことが推察された。これで4種類のAQP分子が小腸に存在することが明らかとなったので、これらに対して解析を進めることとした。
これらのAQPのC末端十数残基のペプチドを調製し、ウサギに免疫して抗AQP抗体を作製した。しかし十分な力価が得られなかったので、プロテインGカラムを用いて抗AQP IgGを分離・濃縮した。得られた抗AQP IgGは十分な力価を示しGST-AQPフュージョンタンパクに対するウエスタンブロッティングの結果も良好であったが、AQPを発現させたアフリカツメガエル卵を用いたウエスタンでは陽性シグナルが得られなかった。この理由として卵上でのAQP発現量が少ない可能性を考え、現在各AQP cDNAを最近市販されたアフリカツメガエル卵用高発現ベクターにリクローニングしている。この高発現系が出来次第免疫沈降法を用いて、コレラ毒素の作用によりAQPのリン酸化が起こるかどうかを調べる予定である。
