Research Abstract

本研究では多数のプロスタグランジン(PC)受容体特異的探索分子を設計・合成し、PGの生物活性の分子機構の解明に成功した。まず、中枢神経系のPGI_2受容体に強く結合する受容体探索分子(15R-TIC)の開発に成功し、脳内に新規プロスタサイクリン受容体(IP_2)が存在することを証明した。また、15R-TICのトリチウム標識体を活用して中枢型IP_2受容体の局所分布の視覚化に成功した。さらに、IP_2受容体タンパク質解析のための光親和性標識リガンドの合成にも成功した。続いて、高速メチル化反応を開発して15R-TICの^<11>C含有PETトレーサーの合成に成功し、生きた赤毛ザルの脳PET画像撮影を行って、IP_2受容体が視床および線条体などの中枢神経系に特異的に分布することを明らかにした。抗腫瘍性△^7ーPGA_1メチルエステルに関する研究では、同PGがサイクリンキナーゼ阻害剤であるp21タンパク質の産生を特異的に誘導し、その結果、細胞周期をG_1期で停止させるという分子機構を明らかにした。また、抗腫瘍性PGによるストレスタンパク質Bipの誘導作用を明らかにし、抗腫瘍性PGの細胞内局在を解析して同PGがERに集積すること、またこの集積がストレスタンパク質BiPの誘導作用と相関していることを明らかにした。また、立体科学に着目した分子設計により、代謝に安定でかつ活性および持続性の高い抗腫瘍性プロスタグランジンを創製することに成功した。さらに、癌細胞のPG耐性機構が膜タンパク質であるGS-Xポンプによる細胞外輸送であることなどを明らかにし、その阻害剤の開発にも成功した。この阻害剤と抗腫瘍性PGとの併用による新たな耐性克服法を確立した。
