Research Abstract |
We investigated the possible mechanism of the conditioned fear stress-induced freezing behavior. In addition, endogenous anxiogenic substances were searched using molecular pharmacological technique. Conditioned fear stress-induced dopamine and serotonin release in the prefrontal cortex but not freezing behavior were markedly inhibited by 7 days i.c.v. treatment with 5-HT_6 receptor mRNA antisense oligonucleotides. These results suggest that 5-HT_6 receptors might be involved in certain anxietic behavior. Furthermore, 5-HT_6 receptor antagonist may have properties for a new type of antianxiety. However, further experiments need to confirm the possibility that serotonin is a candidate of endogenous anxiogenic substance.
