In order to determine which mucin genes and glycosyltransferases genes are expressed in the diseased nasal mucosae, nasal polyps, inferior turbinate mucosae, and maxillary sinus mucosae were obtained from the patients with chronic sinusitis or allergic rhinitis at the time of clinically indicated surgery. Mucin gene expression examined by RT-PCR indicated that MUC1, MUC2, MUC4, MUC5AC,MUC5B and MUC7 are expressed in the human nasal mucosae. Northern blot analysis showed that MUC4, MUC5AC and MUC5B are atrongly expressed and that the expression of MUC2 was weak in the nasal mucosae. Glycosyltransferase expression examined by RT-PCR indicated that four fucosyltransferases and three sialyltransferases were expressed to various degrees in the eleven tissues examined. No disease specific expression was observed either in mucin or glycosyltransferase genes expression. The purpose of this the second project was to investigate the expression of the several MUC genes and glycosyltransferase mRNA by RT-PCR in cultured human nasal epithelial cells, to compare their extent of expression in lipopolysaccharide or human neutrophil elastase treated cells with those in non-treated cells prepared from the same tissue, and to evaluate mucin secretion by ELISA in these cells. The mRNA of MUC1,2,4,5AC,5B,7, fucosyltransferase III,IV,VI,and sialyltransferase 3N,3O,4 were detected and that of MUC3,6, and fucosyltransferase V,VII were not detected. The MUC mRNA levels tended to increase in lipopolysaccharide and elastase treated groups, and the mRNA expression of glycosyltransferases and mucin secretion was increased in lipopolysaccharide with serum and elastase treated groups. The results suggest that MUC protein expression pattern may not be much altered but the messages of glycosyltransferases may be altered in response to lipopolysaccharide or elastase, and these changes could result in abnormal mucin secretion.
