Research Abstract

Thermosensitive liposomes are microscopic vesicles that are designed to release their contained drugs effectively in response to hyperthermia. Previously we revealed the combination of thermosensitive liposome containing CDDP and local barin heating showed remarkable antitumor effects against rat malignant gliomas (Int J Hyperthermia : 157-165,1996. J Neurosurg : 180-184, 1996). A new thermosensitive liposome was prepared considering this drug delivery system as clinical use. While a transition temperature of the conventional liposomes have been 42 or 43゚CC to enhance the synergistic effect of hyperthermia and antitumor drugs, the area above 42゚CC was localized in a narrow portion of human glioma around the RF antenna. But the area of 40゚CC extended up to the wide portion of it. So we prepared thermosensitive liposome which can release adriamycin (ADR) above 40゚CC.This liposome showed a significantly great antitumor effect against rat gliomas at 40゚CC.This procedure 1) contributes to the synergistic effect of hyperthermia and antitumor drugs in the regions above 42゚CC, 2) helps to deliver the antitumor drugs more widely to glioma with the temperature of 40゚CC, 3) gives a good chance to many antitumor drugs, such as ADR, which have not been used because they were blocked by BBB despite of their greater antitumor effects.
