Sox17のdimer形成能
以前の研究成果から、SELEX法により、2copyのAACAA下配列が認められ、Sox17が、cooperative binding またはdimerによりDNA結合する可能性が示唆された。そこで本研究計画において、さらに詳細に検討するため、4bpあいたAACAAT配列のinverte d repeat,H4;片方のAACAAT配列にmutationをいれたH4-M1 ; tandem repeatであるT4;7bpあいたinverted repeatであるH7のプローブを作成し、各プローブに対するSox17蛋白の結合能についてgelshiftにより検討した。ソノ結果、1つのAACAAT配列のみ含むH4-M1やtandem repeatに比べ、inverted repeatにSox17の強い親和性を示すことが明かとなった。さらにmonomer dimerの2種類のDNA-蛋白複合体が確認され、さらに興味深いことに、Sox17は、t-Sox17ともhetero dimerを形成しうることが明かとなった。
