延髄孤束核内のNitric Oxideの脳循環および脊髄循環への役割

Research Abstract

延髄孤束核で一酸化窒素(Nitric oxide,NO)が、synaptic transmissionに関与する物質として働き孤束核内で脳・脊髄循環を調節しているとの仮説を証明するため、ウレタン麻酔下のラットの孤束核にNO発生剤sodium nitroprusside (SNP)を微量注入し脳・脊髄血流量の変化を調べた。1.先ず、脳循環について調べた(Pflugers Arch-Eur J Physiol,432 ; 941-943,1996に発表)。(1)孤束核にSNP微量注入で、刺激と同側の大脳皮質の脳血流量は77±8より57±8 ml/min/100gへと有意に(P<0.01)低下し、脳血管抵抗は有意に上昇した(n=10)。(2)NO合成酵素阻害剤L-NMMAで孤束核を前処置すると、L-glutamateを孤束核へ微量注入した時に生じる脳血管収縮反応が起こらなくなった(n=10)。(3)L-NMMAの異性体でNO産生阻害作用のないD-NMMAで前処置した時には、L-glutamate微量注入による脳血管収縮反応は生じた(n=11)。(4)L-NMMAを単独で孤束核に微量注入した時、脳血流量は変化しなかった(n=9)。(5)孤束核より離れた部位にSNPを微量注入(n=9)又は孤束核に人工髄液を微量注入した時(n=10)脳血流量には変化なかった。2.次に、脊髄循環について調べた(Neurosci Res,25 ; 285-291,1996に発表)。(1)孤束核にSNP微量注入で、頸髄・胸髄・腰髄の血流量は有意に低下し、脊髄血管抵抗は有意に上昇した(n=10)。(2)L-NMMAやD-NMMAで前処置しL-glutamateを孤束核へ微量注入した時や、L-NMMAを単独で孤束核に微量注入した時、脊髄循環の反応は脳循環の反応と同じだった。3.さらに、腎・脾循環について調べた(J Auton Nerv Syst,1997,in press)。孤束核にSNP微量注入で、腎・脾血流量は有意に上昇し、脳・脊髄血流と反対の変化を示した。以上より、孤束核内で発生するNOが、synaptic transmissionに関与する物質として働き孤束核内で脳・脊髄・腎・脾循環を調節している事が示唆された。
