Research Abstract |
The adrenocortical responses of mouse perfused adrenal to ACTH or chemical mediators were examined in various immune deficient genetic mice to explore the relation between the adrenocortical function and immune system. (1) Endogenous platelet-activating factor (PAF) and its receptors may play a role in the ACTH-initiated signaling pathway at the phase after responsiveness to cyclic AMP in normal mice, because continuous infusion of PAF receptor antagonist inhibited corticosterone (B) responses of perfused adrenal to ACTH,forskolin, or dibutyryl cyclic AMP. (2) Its PAF receptors may have less function in nude athymic mice than normal mice, because administration of N-methylcarbamyl PAF,a nonmetabolizable PAF agonist, significantly increased B secretion of perfused adrenal in normal mice, but not in nude mice. (3) The secretory responses of B to 100 nM serotonin in ICR nude mice were one fourth those in ICR normal mice, but the responses to serotonin in WBB6F1 W/Wv (mast-cell-deficient) mice were almost same as those in WBB6F1 +/+ mice. The secretory responses of B to 1 uM prostaglandin D2 in nude or mast cell-deficient mice were one third those in the respective control mice.. (4) In rat perfused adrenal, administration of acidic fibroblast growth factor (aFGF) at 1 or 10 nM significantly increased B secretion, while administration of aFGF into third cerebral ventricle activated hypothalamo-pituitary-adrenocortical axis via CRF release in the brain.. PAF and its receptor in ACTH-initiated signaling pathway may play a key role in the relation between the adrenocortical function and immune system. Further investigation for the adrenocortical function in various immune deficient mice may be useful to make clear causes of allergy and autoimmune diseases.
