Research Abstract |
1) The presence of protein translated from the tumor inhibitory gene p53 in human synivial tissues has been investigated by immunohistochemistry. P53 protein was detected in 16 out of 20 rheumatoid arthritis (RA) synovial tissues. Accumulation of p53 protein was related to the activity of RA synovitis. P53 protein was not detected in osteoarthritis or traumatic synovial tissues. 2) Cultured synovial cells were investigated for the presence of p53 protein by immunofluorescence. P53 protein was found in 3-5% of RA synovial cells but not in traumatic synovial cells. 3) Cell lysate derived from ^<35>S-labelled cultured synovial cells were immuno-precipitated by an anti-p53 protein monoclonal antibody. P53 protein was detected more strongly in RA synovial cells compared to traumatic synovial cells or tumor cell line. In RA synovial cells, an extra band near p53 protein was also detected, suggesting the presence of p53-binding protein. 4) PCR-SSCP was performed using DNA isolated from cultured synovial cells from RA patients. However, there were no apparent comformational changes in these DNA.Also no mutation was detected in p53-mRNA derived from RA synovial cells.
