Research Abstract |
1.We have reported that digestion of glycosaminoglycans by chondroitin ABC lyase enhance hybirdization signals in in situ hybridization techniques. (Histochemical Journal, 1997) 2. Gene expression of matrix metalloproteinase (MMP) -1, -2 and -3 was observed in the stromal cell of basal cell carcinoma (BCC) with tumor cell nests in the dermis. But no expression was found in the superficial type of (BCC). 3.MMP-1 mRNA was detected in tumor cells and/or in stromal cells in all cases of squamous (SCC) cell carcinoma, 4 of 6 actinic kertosis (AK) adjacent to SCC and 4 of 16 AKs. MMP-2 and MMP-3 mRNAs were detected in SCC but not in AK. 4.The expression of MMP-3 was correlated with that of MMP-1 localized at the tumor mass and stroma of the invasive area, while MMP-2 mRNA was detcted widely throughout the stroma independent of MMP-1 expression in SCC. 5.Our results indicated that the expression of MMP 1, -2 and -3 showed different localization patterns, suggesting a unique role of each MMP in tumor progression. Moreover, MMP-1 expression could be an early event in the development of SCC,and AK demonstrating MMP-1 mRNA might be in a more advanced dysplastic state, progressing to SCC.
