Research Abstract |
1. The corre1ations between the polymorphism of beta3 adrenergic receptor(AR) gene(trp64Arg) and various clinical manifestations were studied in NIDDM patients. The prevalences of wild type of beta3 AR gene (TT), Trp64Arg heterozygotes (TA) and homozygotes (AA) were 69%, 25% and 6%, respectively. There was no difference in BMI, max BMI in the past and onset age of diabetes among 3 groups. There was no difference in serum leptin level and PAI-1 activity among 3 groups. 2. The contribution of the beta3 AR gene polymorphism to body weight gain during pregnancy was studied in NIDDM women. Excess weight gain during pregnancy, as defined by maximum BMI during pregnancy minus basal BMI before pregnancy exceeding 5, was observed in 12.2% of the wild type (TT), 19.2% of heterozygotes and 28.6% of homozygotes, suggesting that homozygous (AA) subjects with NIDDM tended to show excess weight gain during pregnancy. 3. The activities of tyrosin kinase inhibitor PC-1 in cultured dermal fibroblasts obtained from NIDDM patients were compared to those of control subjects. The PC-1 activity of the NIDDM patients was 85.2*33.lnmol/mg/min (n=17, mean*SD) and significantly higher than that of control (46.2*12.7, n=7). In additions, the PC-1 activity of a patient with Werner's syndrome with insulin resistance was found to be increased.
