Research Abstract |
The aim of this research project is to identify new genes encoding the proteins important for cell growth, among the cell surface molecules associated with homotypic aggregation. We isolated mRNA from CD34 positive cell lines (KG-1 etc) and CD34 positive hematopoietic stem cells of the patients with hematological malignancy, and generated cDNA using oligo dT primer and randam primer. Several cDNA librarys were constructed from the cDNA and expression vector. These cDNA librarys were tranfered into the COS-7 cells by electroporation, and the cells that reacted with CD34 positive cells, were concentrated by the modified panning method. Two cDNA fragments were isolated from cDNA library of KG-1 cell using this system. The sequence analysis revieled that one of the cDNA fragments is identical to the gene of CD54 (ICAM-1 : intercellular adhesioin melecule 1). The other cDNA fragment will analysed succesively. We will try to isolate new gene associated with homotypic aggregation using different cDNA librarys.
