Research Abstract |
Usefulness of cytokeratin (CK) 20 mRNA as a target of reverse-transcriptase polymerase chain reaction (RT-PCR) was examined in the detection of gynecologic cancer micrometastases in pelvic lymph nodes (LNs). RT-PCR amplification of CK 20 nRNA was conducted using total RNA samples. RT-PCR products were stained with ethidium bromide and anlyzed by agarose gel electrophoresis. The detection sensitivity of CK 20 RT-PCR was 1/10^5 (cancer cell/ LN cells). One hundred and seventy two LNs were obtained from 37 patients with primary gynecologic cancers (18 cervical, 10 endometrial, and 9 ovarian), and metastases in each LN were investigated by histological examination (hematoxylin and eosin sections) and RT-PCR method. CK20 expression was detected in 47% of the tumor tissues examined by the 1st RT-PCR and in 100% by the 2nd (heminested) RT-PCR.CK 20 mRNA was detected in all of the primary cancer tissues. In all the 5 LNs, which were histologiclly metastases-positive, CK20 mRNA was detected by RT-PCR.Of the 167 histologically negative LNs, 4 LNs were found to express CK 20 mRNA,indicating the presence of micrometastases which could be detected by RT-PCR but not by histological examination. These results demonstrate the usefulness of CK 20 RT-PCR in the detection by gynecologic cancer micrometastases in LNs.
