Research Abstract

ラット尾動脈からのプリン物質(ATP,ADP,AMP,adenosine)の遊離
1.高感度フォトンカウンティングカメラ・画像解析装置を用いて検討した結果、ラット尾動脈標本において、ノルアドレナリン(NA)および機械的(伸展)刺激はプリン物質の著明な遊離を惹起し、NAによる遊離は内皮除去により消失した。
2.尾動脈の灌流標本において、NAはプリン物質の著明な遊離を惹起したが、流速の増加、即ちshear stressは有意なプリン物質遊離作用を示さなかった。
NO遊離機構と内因性ATPとの関係に関する検討
1.一酸化窒素(NO)合成酵素阻害薬であるL-NAME(ニトロアルギニンメチルエステル)は尾動脈リング状標本においてNAの収縮反応には影響しなかった。
2.L-NAMEは尾動脈灌流標本においてNAの灌流圧上昇作用を促進した。
3.L-NAMEは尾動脈灌流標本において流速増加による灌流圧の上昇には影響しなかった。
4.L-NAMEは尾動脈灌流標本において、ATP存在下で流速増加による灌流圧の上昇を促進した。
以上の結果より、ラット尾動脈において、1)プリン物質はNAおよび伸展刺激により遊離されること、2)shear stressはプリン物質の遊離を惹起しないこと、3)NAはNOの遊離を惹起しないこと、3)shear stressはNA存在下でNO遊離作用示すこと、2)shear stressはATP存在下でNO遊離作用を示すこと、などが明らかにされ、shear stressとATPの双方がNO遊離に重要な役割を果たしている可能性が示唆された。
