Research Abstract

1. Study of serum concentrations of Free or Protein-bound elements
Sera from healthy persons and sera from hemodialysis patients before and after dialysis were filtered through an ultrafiltration membrane (molecular weight cut-off =10,000). The elemental concentrations in the serum and its ultrafiltration samples were measured by multi -channel ICP-AES.The percentage of each elemental content in the ultrafiltration sample for the content in the serum was as follows : 62-67% for Mg, 51-52% for Ca, about 99% for P and about 90% for K and Na. The percentage may be considered to be the percentage in the protein-unbound form of the element in serum. The serum Si level for hemodialysis patients is higher than that for the healthy persons. The Si percentage in the protein-unbound form hemodialysis patients was 51 % before dialysis and 29% after dialysis. The concentrations of Mg in not only serum but also ultrafiltration sample from hemodialysis patients were significantly higher than those fo … More r healthy persons (p < 0.001). This difference seems to originate for the high of the Mg concentration in the dialysate solution used recently further than Mg levels of healthy subject.
Sera from patients with various diseases (liver disease, renal disease and diabetes mellitus) were also assayed in a similar way to that mentioned above. Both the serum level and the percentage in the unbound form of many of these elements differed significantly between disease groups and the healthy controls. This suggests that these parameters may be associated with these diseases.
2. Fractionation of serum proteins by gel filtration
We attempted to identify molecular weight fractions which contain protein-bound elements. For this purpose, we conducted a basic experiment to fractionate serum proteins according to their molecular weights, by means of gel filtration of pooled sera. Gel filtration using Sephacryl S-200 HR yielded 3 peaks of serum protein. The estimated molecular weight was about 700,000 for the first peak, 170,000 for the second peak and 70,000 for the third, Gel filtered serum fractions, harvested with a fraction collector, were concentrated and electrophoresed to identify peak elements. The concentration of each element in each fraction will be studied in the near future.
3. Study of serum Si levels
In healthy persons, serum Si levels are too low to allow quautification using ICP-AES.We therefore attempted to measure Si levels using atomic absorption spectrometry and obtained favorable results. The results were compared with those yielded by ICP-AES using the standard addition method. Less
