Research Abstract

1)Among 7 yellow colors (carotene, curcumin, carthamus yellow, gardenia yellow, monascus yellow, yellow No.4 and 5), carotene, curcumin, carthamus and gardenia yellow inhibited both A-23187- and thrombin-induced thromboxane B_2(TXB_2) synthesis in vitro. In a pretreatment experiment, all colors except yellow No.4 affected both agonist-induced TXB_2 synthesis. These effectsrevealed to be reversible in case of yellow No.5 and gardenia yellow.
In an ex vivo experiment, both artificial yellow colors (No.4 and 5) inhibited A-23187-induced TXB_2 synthesis and stimulated that which was thrombin induced.
2)l blue colors(gardenia blue, spirulina, blue No.1,2 and green No.3) used in the experiment inhibited thrombin-induced TXB_2 synthesis in vitro, but, blue No.2 and green No.3 had no effect ex vivo. In an aggregation experiment, all colors except blue No.2 inhibited or lagged both agnist-induced platelet aggregation.
3)Among brown colors (caramel, onion, kaoliang and cocoa colors), caramel, onion and kaoliang colors had an inhibitory effect on both agonist-induced TXB_2 synthesis. These 3 colors lagged A-23187-induced platelet aggregation, too.
Each of 6 preservatives(sodium dehydroacetate, Sodium benzoate, potassium sorbate, sodium propionate, butyl-p-hydroxybenzoate and hinokitiol, which is certified in Japan, used in the experiment inhibited thrombin-induced TXB_2 synthesis in vitro at the concentration of acceptable daily intake (ADI)level. The inhibitory effect of butyl-p-hydroxybenzoate was irreversible. In an ex vivo experiment, sodium dehydroacetate and hinokitiol inhibited both agonist-induced TXB_2 synthesis, and the latter inhibited platelet aggregation.
