Research Abstract

魚類の光受容器官である松果体と網膜において合成されるホルモン,メラトニンは環境の明暗と生物時計の制御を受けて合成され,様々な生体リズムの制御に関与していると考えられている.本研究においては,魚類におけるメラトニンの生理機能を解明するための基礎実験として,メラトニン受容体の分布と機能について解析することを試みた.
1.メラトニン受容体の分布オートラジオグラフィーによる解析.2-[^<125>I]iodomelatoninをリガンドとしてin vitroオートラジオグラフィーによりキンギョ脳内メラトニン受容体の局在を解析した結果,メラトニン受容体は視蓋,小脳分子層,視床下部,網膜に高密度に存在することが明らかになった.
2.メラトニン受容体遺伝子のクローニング.これまでに報告された脊椎動物のメラトニン受容体遺伝子の塩基配列をもとにdegenerate primerを作成し,キンギョ脳total RNAを鋳型にPCR法によりcDNAクローニングを行った.その結果,4種のメラトニン受容体をコードすると思われる塩基配列を増幅することができた.これらのクローンは脊椎動物のメラトニン受容体の3番目の膜貫通部位の3′-側から7番目の膜貫通部位に相当するもので,2つはMel_<1a>サブタイプを,他の2つはMel_<1b>サブタイプをコードしていることがhomology searchの結果より明らかになった.全長をコードする遺伝子のクローニングを現在行っている.
