造血器腫瘍の増殖と分化におけるInterleukin-10の役割

Research Abstract

小児における造血器腫瘍細胞とそれらの主たる増殖の場である骨髄のstroma細胞の両面からIL-10の造血器腫瘍の増殖,分化に対する役割について調べた.
【方法と結果】白血病細胞株17種,白血病及び悪性リンパ種の臨床検体70例の腫瘍細胞を用いてRTPCR法によりIL-10,IL-10receptor(R)の発現を調べたところ,それぞれ1種,9例,2種,14例で発現が見られた.また造血器腫瘍細胞の増殖部位である骨髄stroma細胞ではIL-10のみ発現がみられた.IL-10発現例についてはELISA法にても細胞からのIL-10の分泌を確認し,これはTPA刺激により増強された.
次に,IL-10,IL-10Rが発現している一部の細胞株を用いて,IL-10,抗IL-10中和抗体と共培養したときの腫瘍細胞の増強,分化,及びstroma細胞への接着(PKH26蛍光色素による生体染色)について検討したがIL-10は増殖,分化に直接的な影響は及ぼさず,stroma細胞への接着に関してはIL-10にて抑制,抗IL-10中和抗体添加にて増強を認めた.
【結論】IL-10は腫瘍細胞の増殖,分化には関与を認めず,stroma細胞との接着に影響を及ぼすと考えられた.
