腸管由来エンドトキシンの脳・腸・肝・免疫系連関における生理活性物質としての意義

Research Abstract

本研究の目的は、脳・腸・肝・免疫系連関における腸管由来エンドトキシン(LPS)の役割を、(1)血中微量LPSの高感度測定法の確立、(2)非炎症性ストレスに伴う肝臓IL-6産生における腸管由来のLPSの意義の解明、(3)肝におけるIL-6産生細胞の同定を解明することである。
前年度は、(1)無麻酔ラットの血中の微量LPSの高感度(1pg/ml)測定法を確立し、(2)ラットを拘束すると、肝門脈血中にLPSが静脈血よりも大きく増えること、(3)PolymixineBなどでLPSを生体内で中和すると、拘束ストレス時の末梢血IL-6増加反応は、消失することなどが判明した。本年度は、さらに、(1)回腸にカテーテルを慢性留置したラットを用い、FITCでラベルしたLPS(1mg/5ml)を回腸内へ注入し、30分後に1時間の拘束ストレスを加えると、肝臓クッパー(K)細胞及び類洞内皮細胞にFITCが取り込まれており、蛍光度は非ストレスラットのそれの5.8倍であった。(2)さらにFITC陽性細胞の約80%は抗IL-6抗体で染色された。(3)拘束ストレスにより、競合的RT-PCRでみたIL-6mRNAが肝臓で増え、脾臓では変化しない。拘束後、肝臓K細胞及び類洞内皮細胞が抗IL-6抗体で染色された。(4)フットショックやケージ交換ストレスにより肝門脈血IL-6が増加し、その反応もLPS生体内中和により消失することなどを見出した。以上より、軽い非炎症ストレス時に、脳からのストレス信号(恐らく自律神経を介する)が腸管粘膜バリアーに影響し、腸管細菌叢からBacterial Translocationを起こし、体内へ侵入したLPSが腸管及び腸管膜リンパ組織や肝K細胞と類洞内皮細胞に作用し、IL-6を産生させること及び脳・腸・肝・免疫系軸において腸管細菌叢由来のLPSが中心的役割を果たすことが明らかになった。
