Research Abstract

心房性ナトリウム利尿ペプチド(ANP)の発見に引き続き、BNPやCNPなどのアイソペプチドが発見されたように多くの生理活性ペプチドにはいくつかのアイソペプチドの存在することが知られている。我々が最近発見した新しい降圧ペプチドの“アドレノメデュリン(AM)"や“PAMP"もアイソペプチドが存在する可能性が高い。本研究では、AMやPAMPのアイソペプチドの系統的検索を行い、さらなる新規降圧ペプチドの発見を目指した。
我々は、AMの降圧活性に必須であるAMのリング構造やC末アミド構造を認識する各種のRIAおよびPAMPの生理活性に必要なc末近傍のRIAを確立し、RIAとHPLCを用いて、ブタ副腎髄質ペプチド分画の、免疫活性の定性を行った。副腎髄質には今までに知られていた20個のアミノ酸よりなるPAMP[1-20]以外に別の免疫活性が認められ、この免疫活性を単離構造決定し、降圧活性を有したPAMP[9-20](PAMP-12)であることを明らかにした。PAMPの生合成機序として、生体内ではPAMP[1-20]以外にPAMP[9-20]が新たな降圧ペプチドとして生合成されていることが示唆され興味深い。
さらに、AMを発見したアッセイ法(血小板cAMP増加作用)を用い、ヒト褐色細胞腫SP-II分画(弱塩基性ペプチド分画)より、降圧活性を有したPHM(peptide histidine methionine)を単離した。
以上のように降圧ペプチドの発見のため、AMとPAMPのアイソペプチドの系統的検索を行ってきた。免疫活性や血小板cAMP増加活性をもちいた検討では、既知のペプチドの溶出する位置とは異なる位置に多くの活性が認められ、今後これらの分画よりAMとPAMPに続く新しい血管作動ペプチドの発見が期待される。
