Research Abstract

1.新しいKIFsモーターの探索とその機能の解析
大脳、小脳、海馬、網膜、腎臓、小腸等のcDNAライブラリーより、ATP結合共通配列及び微小管結合共通配列を用いて新しいKIFsを探索し、現在までにKIF1C,6,7,8,9,10,11,12,13A,13B,14,15,16A,16B,17の16個の新しい遺伝子を同定(Nakagawa et al.,PNAS 1997)、Northern blottingの結果をもとにKIF1C,12,13A,13B,14,16A,16B,17のクローニングと解析を行い、その機能の分子細胞生物学的解析を行っている。その内、神経細胞内でのKIF3の機能については、培養神経細胞にKIF3のモーター活性を阻止する抗体を微量注入することにより神経軸索の伸長が抑制され、KIF3がKAP3-αFodrinの結合を介してFodrinに結合した小胞を輸送していることが判明した(Takeda et al.JCB 2000)。新しいKIF17が神経特異に発現し、しかも樹状突起でKIF17-mLin10-mLin2-mLin7-NR2Bの結合を介してNMDA受容体を含む小胞を輸送していること(Seto et al.Science 2000)、及びKIF13Aが多種類の細胞でKIF13A-β1adaptin-AP1adaptor complex-mannose 6 phosphate受容体をゴルジ体から形質膜へ輸送していることを明らかにし、モーター蛋白がどのようにcargoを認識するかについて突破口を開いた。
2.KIF1Aモノマーの作動原理の生物物理学的解析
ユニークなモノマー型モーターであるKIF1Aが真に一分子だけで微小管上を一定距離processiveに動く事が可能であるか調べる為、KFI1Aのモーター領域1分子に蛍光標識を一個つけ微小管上を動くか否かを新たに開発した顕微鏡システムを用いて解析した結果KIF1Aモーター領域は1個でprocessiveに微小管上を動く事が証明された(Okada & Hirokawa,Science 1999)。またKIF1Aのk loopとチュブリンのC末E hookの結合が基本的に重要であることが示された(Okada & Hirokawa,PNAS 2000)。
3.急速凍結クライオ電顕を用いたKIF1Aモノマーモーターの … More 3次元構造解析
モノマー(1本足モーター)として存在するユニークなKIF1Aの作動機構を解明するためKIF1Aモーター領域をSf9細胞で精製し微小管に結合させ急速凍結しクライオ電顕で観察した。helical reconstructionを行うことによりKIF1Aモーター領域と微小管複合体の三次元超微細構造が明らかとなりKIF1Aはモーター領域の3ヶ所で微小管と結合している事が明らかとなり、その内の1ヶ所はLoop12のK loopにより形成され重要な働きをしていることが明らかになった(Kikkawa et al.Cell,2000)。
4.Gene targeting法によるKIFs及びMAPs遺伝子欠失マウスの作製とその解析
KIF3B-/-及びKIF3A-/-のmouseもembryonal lethalで内臓(特に心臓)の左右軸における転位がみられ左右軸決定がrandomになっており、これらのマウスの解析の結果、KIF3モーター複合体が胎児初期に形成されるNodeの上皮細胞でその線毛を形成する蛋白複合体を線毛内で輸送しその線毛が時計回りの回転運動をすることにより胎児外液の右から左へのノード流を作りこれが左右決定の最初期現象にとって重要な役割をすることが明らかとなった(Nonaka et al.Cell 1998;Takeda et al.J Cell Biol 1999;Okada et al.Mol Cell 1999).神経特異に発現されるKIF5Cのtargetingを行い、KIF5CがMotor Neuronsの機能と生存に重要な働きをしていることを明らかとした(Kanai et al.,J.Neurosci.2000).現在細胞レベルでの小器官の輸送の解析およびloxPシステムを用いたconditional targetingを行っている。また、細胞の形作りに関わると思われるMAP1BとTauのDouble knock out mouseを作製し解析した結果、MAP1BとTauが相補的に神経の軸索伸長に重要な働きをしていることが明かとなった(Takei et al.JCB 2000)。これに平行してKIF1B,KIF2,KIF4,KIF5B,KIF5C,KIFC2,Brain Dynein等のGene targetingが順調に進行している。MAP1B,MAP2のdouble knock outの解析も順調に進んでいる。
5.遅い軸索輸送の分子機構について
イカの巨大軸索に蛍光標識蛋白を微量注入しその輸送動態を1分子の揺らぎを検索できるFluorescent correlation spectroscopyを用いて解析し、またモーター分子の作動をブロックする抗体の微量注入を組み合わせることにより、チュブリンはオリゴマーとしてキネシンをモーターとして運ばれていることを明らかとした(Terada et al.Cell 2000)。 Less
