Research Abstract

1)LEDボトル(Suzuki et al, 2007)をベースに,パルス幅変調(PWM)法によって,明期,暗期,暗期中断時間,暗期中断開始期および明期・暗期中断の光強度を,それぞれ独立に制御するシステムを開発した.このシステムを用いて,ナミハダニの休眠誘導に暗期中断が及ぼす影響を調査した.その結果,暗期中断によってナミハダニの休眠誘導は阻害され,その積算光強度と休眠率の間に強い相関がみられた.また,カンザワハダニの休眠実験にも使用できることも明らかにした.今後は,本システムをハダニ類以外の微小生物の光周性実験にも応用していきたい.
2)ハダニ類の紫外線適応機構を明らかにするために,強紫外線環境に生息するギリシャ産ナミハダニのUV-B耐性を調査した.その結果,冬期の野外(つくば市)における平均日積算強度と同等のUV-Bを照射した場合,対照区(UV-Bなし)と比較して,孵化率は有意に低下することが明らかになった.今後は,同じシステムを用いて日本産ナミハダニのUV-B耐性を調査し,両系統間に差が見つかれば,生化学・分子生物学的アプローチによって分子機構の比較調査へと研究を展開していきたい.
3)光環境調節によるハダニ類の行動制御を目的とし,微小移動運動補償装置(Sakuma, 2002)を用いて,ナミハダニのフォトキネシスを調査した.その結果,非休眠状態のナミハダニは,UV-Bおよび可視光に対して,それぞれ忌避および誘引行動を示した.今後は,これら行動における作用スペクトルを明らかにし,ナミハダニの行動制御に有効な照明システムを開発していきたい.
