Research Abstract

田中は米国で開かれたFASEB meetingに参加しテロメアの維持機構について情報交換を行い、その後、カリフォルニアのブラックバーン研究室でテロメアの解析法を修得した。それらの情報および方法を用い、in vivoでMre11タンパク質がテロメア領域に結合していることを明らかにし、また、細胞周期におけるテロメアの構造変化に組換え遺伝子が関与していることを明らかにしつつある。太田は組み換え遺伝子Mre11タンパク質がDNA結合能を持つこと、ヌクレアーゼ活性を持つことを明らかにし、雑誌Ce11に発表した。また、mre11の種々の変異株を作成し、ヌクレアーゼ活性が組換え修復機能み重要な役割を果たしていることを明らかにした。田中は太田の作成したmre11の変異株を用いMre11タンパク質のDNA結合領域がテロメアの維持に重要な役割を担うことを明らかにした。
