Research Abstract

本研究は、ヒトのがんの最大の標的である上皮組織について、その形態形成、増殖、分化の制御機構の解析を目的とする。対象とする上皮組織は、表皮ケラチノサイト、子宮頚部扁平上皮、肺胞上皮、胃上皮などである。
1)形態形成; マウス皮膚は胎生13日から16日の間に表皮の多層化、角化、毛のう原基の形成という組織構築の基本構造が作られる。このとき発現する遺伝子をDifferental RNA display 法によって分離した胎盤から組織浸潤関連遺伝子を探索した。
2)増殖制御; ヒト重層扁平上皮の上皮細胞をintegrin β1,integrin β4, EGFR, p75NGFR, bcl-2, BP-180, Ki-67 をマーカーに分画し、有力な幹細胞の候補と考えられるp75NGFR 強陽性でIntegrin β1 陰性細胞サブセットを分離同定した。増殖停止機構の解析、上皮系のアポトーシス機構、パラクリン増殖因子の同定などを進めた。
3)細胞分化; シグナル伝達に関わるCキナーゼ分子種を明らかにし、その遺伝子操作動物を作成した。ケラチノサイトの終末分化の際のG1 arrest はCキナーゼn分子種(PKCn)がcdk-2/cylin E 複合体と結合し、cdk-2活性を阻害、Rb蛋白のリン酸化を抑えるためであることを見いだし、終末分化においてG1停止と分化の誘導が独立したメカニズムによって進行することを明らかにした。
