Research Abstract

私たちはHIV転写制御因子、Tatの細胞内選別輸送とその制御機構およびそれに関与する細胞質因子の同定を含めた分子レベルでの解明を目的として、Tat依存的gpt(guanine phosphoribosyl transferase)遺伝子の発現、すなわち6-チオグアニン感受性を表現型としたヒトT細胞CEM由来細胞株を分離した。Ro-24-7429をはじめ、Tatアンタゴニスト候補物質を用いてこの細胞株の表現型の変化(6-チオグアニン感受性から耐性への変換)を調べたが、現在のところ、顕著な感受性変化への効果は見出されていない。さらに現在、Tat核輸送シグナルを有する合成ペプチドなど他のTatアンタゴニストと報告されている物質についてその阻害効果、すなわち細胞株の有用性を検討している。
