Research Abstract

抗原ペプチドがプロテオゾーム-TAP1.2経路でERに移入される際、ここにhsc70が関与し、ひとつのcircuitが存在すると考えられた。すなわち、プロテオゾームとATP結合型hsc70の複合体に抗原ペプチドが結合し、hsc70自身およびATPase活性を有する他の要因により、抗原ペプチドはADP結合型hsc70と複合体を形成する。これが、TAP1.2と会合し、ここでATPの働きにより抗原ペプチドが解離し、TAP1.2を通じてER内に移入されると考えられる。一方、MeDSG(methyl-deoxyspergualin)はhsc70とTAP1.2の会合そのものを阻止するため、hsc70に結合する抗原ペプチドはERに移入できなくなると考えられる。その結果、MeDSG処理によりMHC class I分子の細胞表面発現低下がおこると考えられた。このことはMeDSGの免疫抑制剤としての作用のひとつの側面を説明するものと考えられた。
