Research Abstract

様々な細胞膜タンパク質の細胞外ドメインが、増殖因子の刺激などに際して切断され、その生理活性が変化することが知られている。この現象はPKCを活性化するTPA等によっても引き起こされ、この過程にPKCを介する細胞内シグナル経路が関わっていることが推測される。しかしながら、これに関わるプロテアーゼの実体さらにはその切断の分子機構は全く不明である。我々はPKCdに特異的に結合するタンパク質として、MDC9と呼ばれる細胞膜貫通型のタンパク質を見いだした。MDC9は細胞外にメタロプロテアーゼの配列、インテグリンに結合する配列等を有すると同時に、細胞内にPKCによるリン酸化コンセンサス配列、SH3ドメインの結合配列等を有するタンパク質であるがその機能は全く不明であった。PKCdはMDC9の細胞質ドメインのリン酸化コンセンサスを含む部位と特異的に結合しリン酸化する事を確認した。更に、PKCdとMDC9との特異的な相互作用の生理的な意義を調べる目的で、TPAにより細胞外ドメインの特異的な切断が起きることが知られている細胞増殖因子、HB-EGFのプロセシングヘの関与を調べた。HB-EGFは通常細胞膜に止まった状態で存在し、細胞接着を介して増殖因子として作用するのに加え、細胞の刺激によりその細胞外ドメインが切断され、遊離型の増殖因子としてパラクラインに作用する事が知られている。PKCdやMDC9の活性型変異体や抑制型変異体を作成し、HB-EGFのプロセシングに対する効果を調べた結果、PKCdとMDC9とは、HB-EGFの刺激依存的なプロセシングに関わっていることを証明した。同様の現象は、多細胞生物の発生過程等を含め普遍的に起きる現象であり、PKCd及びMDC9の作用の普遍性が推測される。
