Research Abstract

血管制御因子である一酸化窒素(NO)やスーパーオキシドが高血圧や動脈硬化の病因に関与する可能性が示唆されているが、その分子論的実体は尚不明である。我々は、個体レベルで血管内皮細胞に特異的に濃縮動員されるSOD(ヘパリン結合性HB-SOD)を開発し、血管局所のNOとスーパーオキシドの不均衡が高血圧や循環病態の重要な要因であることを世界に先駆けて示してきた。また、短寿命と信じられていたNOが生理的低酸素分圧下ではGSHなどと相互作用しながら極めて長時間機能することを明らかにした。本研究は、血管局所のNOとスーパーオキシドのクロストーク、酸素代謝、及びチオール代謝動態を個体レベルで制御し、動脈硬化や高血圧病態における活性酸素の意義を明らかにし、新たな治療法の開発を目的として行われた。
解析の結果、微量のNOでは低酸素下でのみ血管壁でのcGMP産生増加が見られること、およびミトコンドリアのATP合成が可逆的に抑制されることを見いだした。本現象が個体レベルでも起こること、これが虚血により増強されること、および酸素、スーパーオキシドおよびNOの局所的バランスが循環制御因子として作用していることが明らかになった。
動脈硬化モデルのWatanabe兎、及び本体性高血圧ラット(SHR)では、組織のGSH濃度には有為な差が見られないが、その代謝回転量は著明に低下していることが判明した。このことから、両循環病態動物では、血管のみならず、全身組織で酸化ストレスが亢進していることが明らかになった。また、血管および全身組織の酸素ストレスを特異的に制御する事が循環病態改善に重要であることが判明した。
