Research Abstract

EBウイルス(EBV)の上皮細胞への感染を簡便にモニターするために、蛍光蛋白Enhanced green fluorescent protein(EGFP)遺伝子を組み込んだ組み換えEBV(EGFP―EBV)を作製した。このウイルスを用いた検討により、いくつかのヒト上皮細胞株がEBV感染感受性を有することを明らかにした。これらの上皮細胞株は、これまでに知られているEBVレセプター(CD21)をほとんど発現していなかった。さらに抗CD21抗体を用いたプロッキング実験により、上皮細胞への感染がCD21以外の分子をレセプターとして成立することを証明した。
次に、CD21に結合するウイルス側のリガンドgp350/220とヒトIgG1Fc部分との融合蛋白gp350/220-Igを作製した。この融合室白はCD21を発現するB細胞株に強く結合したが、上皮細胞株にはほとんど結合しなかった。一方、他のウイルス粘蜜白gp25とヒトIgG1Fc部分との融合蜜白gp25-Igは、 B細胞株にはほとんど結合せずEBV感染感受性の高い上皮細胞株に強く結合した。この結果から、上皮細胞に発現するEBVレセプターに結合するウイルス側のリガンド分子はgp25であると考えられた。
種々のgp25分子の欠失変異体を作製することにより、gp25分子中の上皮細胞への結合に必要な領域を決定した。その結果、アミノ酸71-100の領域が上皮細胞への結合に必須であることが明らかとなった。この領域と相同性のある蛋白をBLAST homolgysearchにより検索したところ、脂質二重膜の融合に働くsyntaxin分子との相同性が認められた。Gp25分子はsyntaxin分子と同様に、脂質二重膜の融合機構に組み込まれて働く可能性が示された。
