Research Abstract |
(1) We have developed a direct immunofluorescent assay kit for detection of Cryptosporidium oocysts in fecal and water specimens. The kit consists of FITC-labeled monoclonal antibodies (IgM) specific for the oocyst wall, blocking solution, and suspension containing C.parvum oocysts. Since the antibodies are highly reactive to oocysts of cryptosporidia and were not cross-reactive to cysts and oocysts of intestinal protozoan parasites except Cryptosporidium, the kit is useful for routine fecal examination of cryptosporidiosis in man and animals. For detection and identification of oocysts from surface water samples, further improvement is needed because of existence of some cross-reactive algae in river water. (2) The ability of an electrolytic bacteriostatic system (model ABE-75, Konica Corp., Tokyo) to remove and inactivate Cryptosportdium parvum oocysts in water was evaluated. In the experiment, 5 liters of wafer spiked with 4 * 10^7 or 1.4 * 10^8 oocysts was treated with the electrolyzer. More than 99.99999% of the oocysts were removed within 5 h by filtration with the porous cabon graphite, and the infectivity toward mice of oocysts obtained from the porous cabon graphite electrodes was reduced to 4% within this time. (3) Inactivation of Cryptosporidium oocysts in water by ozonation was evaluated. In activation rates derived from animal infectivity were 99.9% by the treatment with 0.5 mg/L ozone for 30 min.
