Research Abstract

本研究の目的は、重原子として希ガス(キセノンとクリプトン)を用い位相決定のための重原子同形置換体を得ることである。そのために我々は、5MPa程度までの圧力に耐えられるガス封入装置を作製した。この装置はキャピラリーと市販の樹脂製T字コネクタを組み合わせた簡単な装置である。
現在までにPFやSPring-8で、希ガスの圧を変化させ、リゾチーム、リボ核酸分解酵素、PCB分解酵素、およびトリプシンの蛋白質結晶に希ガスの導入を試みた。トリプシン以外の蛋白質結晶には、希ガスを導入することができた。しかし、リゾチーム、リボ核酸分解酵素については、単独で位相決定のために利用できるような重原子同形置換体は得られていない。一方、PCB分解酵素については、Spring-8のBL41XUにおいて、1つの結晶から、nativeとキセノン圧2.7MPaのデータを得ることが出来た。この重原子同形置換体は、分解能2.55A、completeness96.6%であった。このnatieveとキセノン同形置換体を処理した結果、パターソン図に明確なピークを得ることが出来た。3箇所のキセノンの座標を用いてプログラムMLPHAREにて重原子サイトの精密化を試みたが、電子密度分布図を解釈することはできなかった。そこで、この精密結果を用いて、プログラムSHARPを用いて更に、重原子サイトの精密化を行った。さらに、プログラムSOLOMONにて溶媒平滑化を行うことにより、分解能2.55Aで、蛋白質分子モデルを構築できる高品質な電子密度分布図を得ることに成功した。すなわち、希ガス単独で位相決定を行うという所期の目的を達成できた。
